【Universal Magnetic Co-IP Kit (商品コード 54002) 】日本語版プロトコールあり
【Nuclear Complex Co-IP Kit (商品コード 54001) 】
タンパク質-タンパク質間結合を解析できるキットです。
従来、核タンパク質複合体の共免疫沈降は核抽出液調製の際に核内タンパク質複合
体が
解離してしまったり、免疫沈降に使用するバッファー中の塩や界面活性剤が多くの
不安定
なタンパク質複合体に影響を及ぼすため難しいとされていましたが、アクティブ・
モティフ社の共免疫沈降キットは、核タンパク質複合体を保持させるための抽出試
薬および
免疫沈降試薬を用いることで、この問題を解決しました。
さらに、Universal Magnetic Co-IP Kitは核タンパク質だけでなく、Whole Cell
Extract
を用いても共免疫沈降ができます。また磁気ビーズを用いて免疫沈降を行うため、
操作が
簡便でバックグラウンドを抑えることもできます(磁気ビーズおよびマグネット
バーは
キット内に含まれます)。
Universal Magnetic Co-IP Kitの特長
● 試料は培養細胞だけでなく、組織も使用できます。
● 核タンパク質複合体または細胞質複合体の共免疫沈降に最適です。
● 磁気ビーズを使用することで, 操作が簡便かつ迅速になるだけでなく,
バックグラウンドを抑えます。
● 核抽出物調製の際に, 核タンパク質複合体のタンパク質間結合を解離させま
せん。
● タンパク質修飾を保持するよう, キットには様々な阻害剤も付属していま
す。
Universal Magnetic Co-IP Kitの簡易手順
1. 培養細胞または組織から、核抽出液（または全細胞抽出液）を調製する。
2. 酵素処理によりDNAの断片化(核抽出液の場合のみ)を行う。
3. 目的タンパク質のうちの1つを認識する抗体と反応させる。
4. 磁気ビーズを加える
5. ウォッシュ後、回収する。
6. 目的タンパク質のうちのもう1つを認識する抗体でウェスタンブロッティング
を行う。
